Position description

Assistant Specialist

Job Description:

The Huebner lab investigates human development and disease using CRISPR tools, multicellular models, and functional genomic screens. The research projects utilize human iPSC-based models and functional genomics to uncover the molecular basis of developmental processes and the origins of cellular transformation. The Specialist will be part of a team that performs complex experiments including genome scale screens, genetic interaction mapping, and genome engineering. The Specialist will assist in the design, execution, and analysis of the following experiments in support of these research projects: (1) targeted and genome-wide pooled genetic screens in human cells, (2) production/testing of lentiviral sgRNA libraries for use in mammalian cells, (3) implementation of cell-based assays including flow cytometry, luciferase reporter detection, and microscopy-based approaches, (4) next-generation sequencing including single cell sequencing and (5) data analysis. The Specialist is expected to demonstrate the capacity to work in a semi-independent manner and as part of a team, as well as disseminate their findings in lab meetings and other public settings.

Required Qualifications:



  • A master's degree in a biology-centered field or a baccalaureate degree with three or more years of direct cancer/stem cell research experience.
  • 2+ years' experience deriving and maintaining mammalian cell lines including human iPSCs and 3D organoids
  • Previous experience using FACS analyzers and sorters
  • Prior experience performing immunofluorescence and confocal microscopy
  • Experience in the execution of CRISPR growth/FACS screens, including guide library preparation and analysis
  • Ability to generate lentivirus and transducing mammalian cells
  • Proficient in molecular cloning techniques such as restriction digests, Gibson cloning, and plasmid design
  • Proficient use of Python, R, Adobe Illustrator, and GraphPad


Preferred Qualifications:



  • Familiarity with stem cell/developmental biology
  • Experience generating genetically engineered cell lines
  • Processing, QC, and alignment of NGS data (bulk RNAseq, scRNA-seq, ATACseq)
