Position description

The UCSF Division of Cardiology/Cardiovascular Research Institute (CVRI) is seeking a highly motivated and skilled full-time Histology Technician to support cutting-edge research on cardiovascular biology using primarily mouse models (embryonic and adult). The successful candidate will play a critical role in the preparation, analysis, and imaging of tissue samples, contributing to groundbreaking discoveries in the field.

Key Responsibilities:

* Tissue Preparation and Sectioning:

o Perform fixation, processing, and embedding of mouse tissues (both embryonic and adult) for histological analysis.

o Section tissues using microtomy (paraffin) and cryotomy (frozen) techniques with high precision.

* Histological and Molecular Staining:

o Execute standard histological stains (e.g., H&E, Masson's trichrome).

o Conduct specialized staining protocols, including immunohistochemistry (IHC), immunofluorescence (IF), and advanced RNA in situ hybridization techniques such as RNAscope.

o Optimize and troubleshoot staining protocols as required.

* Microscopy and Structural Identification:

o Utilize light, fluorescent, and confocal microscopy to analyze stained tissue samples.

o Identify and document key structures of interest, ensuring accuracy and relevance to specific research objectives.

o Collaborate with researchers to confirm findings and provide interpretative input when necessary.

* Spatial Transcriptomics:

o Gain and apply expertise in spatial transcriptomic technologies to map gene expression within tissue samples.

o Prepare and process tissue for spatial transcriptomic experiments, adhering to rigorous quality control standards.

o Collaborate with bioinformatics teams to integrate spatial data into broader research contexts.

* Data Documentation and Quality Control:

o Maintain accurate and detailed records of all sample preparation, staining protocols, and analytical results.

o Ensure consistency and reproducibility of histological processes by adhering to established standard operating procedures (SOPs).

o Participate in the evaluation and implementation of new technologies and methods to enhance histological capabilities.

* Lab Collaboration and Communication:

o Work closely with a multidisciplinary team of researchers, including molecular biologists, physiologists, and bioinformaticians.

o Provide technical support and training to team members as needed.

o Attend and contribute to lab meetings, presenting data and updates on histological findings.

Required qualifications:

* Bachelor's degree in biology, biochemistry, molecular biology, or a related science field OR at least four years of research experience.

* Hands-on experience in tissue preparation, sectioning, and a variety of staining techniques (e.g., RNAscope, IHC, IF).

* Proficiency with microscopy (light, fluorescence, confocal) and identifying tissue structures.

* Familiarity with or willingness to learn spatial transcriptomic techniques.

* Strong attention to detail and excellent organizational skills.

* Ability to work both independently and collaboratively in a dynamic research environment.

Preferred qualifications:

* Experience with mouse models and tissue handling.

* Knowledge of cardiovascular biology or related research fields.

* Competence in troubleshooting and optimizing histological protocols.
